Enzymic lipoxygenation of arachidonic acid: mechanism, inhibition, and role in eicosanoid biosynthesis

چکیده

Studies on the lipoxygenation of arachidonic acid by various enzymes have been carried out using diverse approaches. A mechanistic picture of this reaction has been developed which both correlates known information and assists in the rational design of new lipoxygenase inhibitors. A working hypothesis is advanced for the biosynthesis of prostaglandins in mammalian systems. An entirely different mechanism is proposed for the biosynthesis of prostaglandins and related eicosanoids in marine organisms.

Inhibition of U937 eicosanoid and DNA synthesis by 5,8,11,14-eicosatetraynoic acid, an inhibitor of arachidonic acid metabolism and its partial reversal by leukotriene C4.

When replicating U937 cells were incubated with up to 80 microM concentrations of the in vitro inhibitor of eicosanoid biosynthesis, 5,8,11,14-eicosatetraynoic acid (ETYA), DNA synthesis measured by labeling with [3H]thymidine was inhibited in a concentration-dependent manner. No reduction in cellular viability occurred, as judged by exclusion of trypan blue, unaltered release of 51Cr-labeled p...

The influence of chronic eicosanoid biosynthesis inhibition on life history of the greater waxmoth, Galleria mellonella and its ectoparasitoid, Bracon hebetor.

Eicosanoids are oxygenated metabolites of three C20 polyunsaturated fatty acids, mainly arachidonic acid (AA; 20:4n-6), but also 20:3n-6 and 20:5n-3. Aside from their importance in biomedicine, eicosanoids act in invertebrate biology. Prostaglandins (PGs) influence salt and water transport physiology in insect rectal epithelia and in Malpighian tubules. PGs also influence a few insect behaviors...

Biological and chemical stimulators cause tissue injury. Many epidemiological studies imply that chronic stimulation of tissues leads to cancer. One of the most important type of chronic tissue stimulation criteria is increased activity of the metabolic pathway of arachidonic acid and production of biochemical intermediates. Cyclooxygenase pathway (COX) of arachidonic acid leads to production o...

Enhanced arachidonic acid lipoxygenation by K562 cells stimulated with 12-O-tetradecanoylphorbol-13-acetate.

The arachidonic acid lipoxygenase products released into culture supernatants by the human myeloid cell line K562 were quantitated by high-performance liquid chromatography. During 2 hr of incubation, K562 cells spontaneously released a mean of 9 ng of 15-monohydroxyeicosatetraenoic acid, 35 ng of 5-monohydroxyeicosatetraenoic acid, and 87 ng of a partially resolved mixture of 11- and 12-monohy...

The entomopathogenic bacterium, Xenorhabdus nematophila, impairs hemocytic immunity by inhibition of eicosanoid biosynthesis in adult crickets, Gryllus Wrmus

The bacterium, Xenorhabdus nematophila (Poinar and Thomas), is an obligate symbiont of nematodes in the genus Steinernema and a lethal insect pathogen. We investigated the hypothesis that one aspect of the bacterial virulence is the ability of X. nematophila to severely impair host insect cellular immune reactions to infection by inhibiting eicosanoid biosynthesis in the adult male cricket, Gry...
